Sumoylation 10x Reaction Buffer

Description:
Each tube contains 200 µl of sumoylation 10x reaction buffer (200 mM Hepes pH 7.5, 50mM MgCl2, 20mM ATP).
Assay Conditions:
Sumoylation assays are carried out in a final volume of 20µl in reaction condition (20 mM Hepes pH 7.5, 5mM MgCl2, 2mM ATP). We would recommend to add fresh 2mM ATP to be sure that sufficient energy is supplied.
Sumoylation Protocol:
1. Prepare the suitable purified substrate protein.
2. In each reaction, add 1 µl UBC9 (E2) to substrate first, then 1 µl Sumo-1, 2 µl 10x reaction buffer, 1 µl Aos1/Uba2 (E1). Finally, add H2O to bring up to 20 µl (we would recommend to add fresh 2mM ATP to be sure that sufficient energy is supplied).
3. The best reaction concentration of proteins is as following:
E I: 7.5µg/ml
E II: 50µg/ml
SUMO-1: 50µg/ml
